  10.  AB    OBJECTIVE: To provide a highly sensitive assay by simultaneous detection
  11.        of anti ENV HIV-1 and/or HIV-2 antibody and/or GAG antigen with
  12.        characterization of each reactive analyte. METHODS: Well documented
  13.        serial bleeds of HIV-1 seroconverters were tested for p24 antigen, gp41
  14.        and HIV2 gp36 antibodies with a unitary membrane test strip containing
  15.        specific bands for each analyte to be detected. This assay, called
  16.        DECISCAN HIV Ab/Ag, is based on a sandwich ELISA format using HIV1 and
  17.        HIV2 ENV antigens (synthetic peptides and recombinant glycoprotein) for
  18.        antibody detection and monoclonal antibodies for p24 detection. RESULTS:
  19.        In all seroconverters tested, HIV-1 antibody detection bands were
  20.        reactive in the same bleeds as in 3rd generation antibody assays. In
  21.        three patients, p24 was detected in at least one bleed prior to antibody
  22.        detection. Three other seroconversions became antibody and antigen
  23.        positive in the same bleed. One seroconverters became antibody reactive
  24.        without antigen detection. No reactivity was observed in 200 HIV
  25.        antibody negative samples from normal blood donors. DISCUSSION AND
  26.        CONCLUSION: The new DECISCAN HIV Ab/Ag supplemental assay allows an
  27.        early diagnosis of HIV seroconverters based on highly sensitive anti ENV
  28.        HIV antibody detection improved by simultaneous p24 antigen detection.
  29.        This assay gives useful additonal data for patients with low antibody
  30.        reactivity or WB indeterminate results.
